Product Descripition

This product is a high-fidelity DNA polymerase premix formulated with a specialized recipe, containing essential components such as dNTPs and a buffer system. It ensures efficient amplification of long DNA fragments with exceptional fidelity, enabling ultra-fast amplification at a rate of just 5 seconds per kilobase. The premix is highly versatile, performing well on standard templates, challenging high-GC systems (whether involving primers or templates), and even templates containing uracil. Key to its performance is the introduction of an optimized Hot Start high-fidelity DNA polymerase, whose unique design and proprietary additives enhance its affinity for template DNA. Combined with robust 3’→5’ exonuclease activity, this enzyme guarantees unparalleled accuracy during DNA amplification. At the same time, the product delivers outstanding advantages, including rapid amplification speed, superior sensitivity, and excellent fidelity. This product comes as a dye-containing 2X Mix, meaning all you need to do during your experiment is add your template and primers to initiate amplification. After the reaction is complete, there’s no need to add an extra Loading Buffer for visualization—simply proceed directly to sample loading for quick downstream analysis. Plus, thanks to its streamlined workflow, you can skip traditional purification steps altogether, significantly boosting experimental efficiency.

This product is suitable for gene cloning and amplification of complex DNA templates (such as high-GC templates or uracil-containing templates).

Exp erimental case

1.Demonstration of amplification performance

（1）Adapt to amplify different lengths of target fragments

Using 100ng of Human gDNA as the template, the target fragments with lengths ranging from 0.5 kb to 10 kb were amplified respectively using 2X GeniuSTAR HiFi Fast PCR Master Mix (Dye plus) (Code: P0017), with the extension speed set at 5 sec/kb. Good amplification results could be obtained in all cases.

M：DL15000 Marker；From lane 1 to lane 8：the target fragments are 0.5 kb, 1 kb, 2 kb, 3 kb, 4 kb, 6 kb, 8 kb, and 10 kb respectively

（2）High sensitivity

Using λDNA as the template, with template addition amounts ranging from 10pg to 0.1fg, the reaction was carried out using 2X GeniuSTAR HiFi Fast PCR Master Mix (Dye plus) (Code: P0017). The results showed that the product had good sensitivity and could stably amplify as low as 100fg.

M：DL15000 Marker；From lane 1 to lane 6： the template amounts are 10 pg, 1 pg, 100 fg, 10 fg, 1 fg, and 0.1 fg respectively; Lane 7: uses dH₂O as the negative control.

（3）Good stability

2X GeniuSTAR HiFi Fast PCR Master Mix (Dye plus) (Code: P0017) was treated at 25℃and 37℃ respectively, and the product that was normally stored was used as the control. The amplification was performed using a high GC template (GC = 74%). The results showed that both the accelerated preservation samples and the control could stably amplify the template.

Lane 1 and Lane 6: The products are stored normally at the recommended temperature;

Lane 2 to Lane 5: The products are placed at 25℃ for 2 days, 4 days, 6 days, and 8 days respectively; 

Lane 7 to Lane 10: The products are placed at 37℃ for 18 hours, 36 hours, 54 hours, and 72 hours respectively.

2.Comparison of Enzymes for NGS

(1)The yield and fragment length of 200bp fragmented DNA libraries

The results show: 

The yield of the libraries：The yield of T4 (U&G P0016) is comparable to that of product C (the corresponding product from the leading company),the yield of product C (the corresponding product for the leading company) is slightly higher than that of T3 ( U&G P0017 ).

Fragment length：The fragment lengths of T3 ( U&G P0017 ) and T4 ( U&G P0016 ) are comparable to that of product C ( the corresponding product from the leading company .

(2)The yield and fragment length of 300bp fragmented DNA libraries

The results show: 

The yield of the library：The yield of T4 (U&G P0016) is significantly higher than that of the other samples.

The fragment length: there is no significant difference among the 6 samples.

3.Comparison of sequencing data quality of libraries

The sequencing data quality of the NGS libraries amplified by U&G P0016 is basically equivalent to that of the control ( the corresponding product from the leading company ).
